15d-Deoxy-Δ12,14-prostaglandin J2 modulates collagen type I synthesis in human hepatic stellate cells by inducing oxidative stress

摘要: 15 deoxy-Delta(12,14)-prostaglandin(2) (15d-PGJ(2)) is known to inhibit the proliferation of hepatic stellate cells (HSCs), major cellular components that cause fibrosis, in vitro. It also induces oxidative stress, which results myofibroblast death. On other hand, stress generally HSC and collagen synthesis vitro, liver fibrogenesis vivo. In this study, we evaluated effects 15d-PGJ(2) at various concentrations on viability HSCs. increased intracellular reactive oxygen species (ROS), reduced human HSCs 5 microM by inducing apoptotic cell addition, antioxidants alpha-tocopherol N-acetylcysteine (NAC) blocked 15d-PGJ(2)-induced Collagen I was 1.5-fold 0.5 treatment, but 30% control level 10 15d-PGJ(2), NAC pretreatment prevented these changes production 15d-PGJ(2). We conclude may either induce or prevent depending its concentration.
