Peroxisome Proliferator-Activated Receptor α Activation during Pregnancy Severely Impairs Mammary Lobuloalveolar Development in Mice

摘要: To identify the potential functions of peroxisome proliferator-activated receptor alpha (PPARα) in skin development, transgenic mice were generated to target constitutively activated PPARα (VP16PPARα) stratified epithelia by use keratin K5 promoter. In addition marked alterations epidermal had a severe defect lactation during pregnancy resulting 100% pup mortality. this study, alteration mammary gland development these was investigated. The results showed that expression VP16PPARα transgene resulted impaired lobuloalveoli, which is associated with reduced proliferation and increased apoptosis epithelia. Mammary from also significant reduction β-catenin down-regulation one its genes, cyclin D1, thought be required for lobuloalveolar development. Furthermore, upon ligand treatment, similar effects on observed wild-type mice, but not PPARα-null mice. These findings suggest activation has influence
