Monoclonal antibodies against O and K antigens of uropathogenic Escherichia coli O4 : K12 : H− as opsonins

摘要: Monoclonal antibodies of subclasses IgG1 and IgG2b specific for the O4 antigen Escherichia coli 20025 (O4 : K12 H−) capsular polysaccharide same strain (IgM) were obtained with hybridoma technique using spleen cells from Balb/c mice, immunized a crude bacterial extract, Sp2/O-Ag8 myeloma cells. The anti-O4 reacted exclusively lipopolysaccharide not those serologically O-cross reactive E. coli. anti-K12 recognized as epitope (part of) KDO moiety polysaccharide. Not only anti-K12, but also effectively phagoopsonized encapsulated 20025. opsonized bacteria killed in subsequent vitro phagocytosis by human leokocytes presence serum complement.
