Accumulation of mutations in DNA gyrase and topoisomerase IV genes contributes to fluoroquinolone resistance in Vibrio cholerae O139 strains.

摘要: High resistance rates to nalidixic acid (NAL) in Vibrio cholerae serogroup O139 strains have been found, and ciprofloxacin (CIP) is also observed. In this study, mutations within the quinolone-resistance determining regions (QRDRs) of DNA gyrase topoisomerase IV from NAL-resistant were analysed. The predominant mutation profile was S83I GyrA combination with S85L ParC. addition, substitutions D87N D420N ParE ParC as well P439S found CIP-resistant strains. A series site-directed mutants comprising D87 GyrA, D420 P439 constructed a wild-type V. strain carrying common Introduction increased CIP minimum inhibitory concentration (MIC) mutant by nearly 4-fold compared initial strain. second introduction further significantly MIC ca. 23-fold 17-fold. Therefore, it concluded that emergence or dramatically induces fluoroquinolones O139, accumulation multiple QRDRs confers significant cholerae.
