Reduction of Amyloid Angiopathy and Aβ Plaque Burden after Enriched Housing in TgCRND8 Mice

摘要: Diversity and intensity of intellectual physical activities seem to have an inverse relationship with the extent cognitive decline in Alzheimer's disease (AD). To study interaction between active lifestyle AD pathology, female TgCRND8 mice carrying human APPswe+ind were transferred into enriched housing. Four months continuous diversified environmental stimulation resulted a significant reduction β-amyloid (Aβ) plaques lower amyloid angiopathy. Neither precursor protein (APP) mRNA/protein levels nor level carboxy-terminal fragments APP soluble Aβ content differed both groups, making alterations expression or processing unlikely as cause reduced deposition. Moreover, DNA microarray analysis revealed simultaneous down-regulation proinflammatory genes well up-regulation molecules involved anti-inflammatory processes, proteasomal degradation, cholesterol binding, possibly explaining burden by aggregation enhanced clearance Aβ. Additionally, immunoblotting against F4/80 antigen morphometric microglia (Mac-3) significantly elevated microgliosis brains, which suggests increased phagocytosis. In summary, this demonstrates that environment interacts pathology at dif-ferent levels.
