摘要: In order to reduce the time required for development and production of viral vaccines, host cell lines should be available as expression systems vaccines against groups pathogens. A selection was compared their initial feasibility system replication polioviruses, influenza viruses respiratory syncytial virus (wild type strain A2). Six adherent (Vero, HEK-293, MRC-5, CHO-K1, BHK-21 c13, MDCK) six single suspension (CAP, AGE1.CR.HS, sCHO-K1, c13 2p, MDCK SFS) were studied ability propagate viruses. First, maximum densities determined. Second, receptor polarization regarding distribution eight different monitored using flow cytometry immunocytochemistry. Organization actin cytoskeleton by transfection cells with Lifeact™, a construct coding actin-EGFP. Finally, produce progeny assayed each line. The results suggest that grown on serum free medium are best candidates serve replication.
