Identification of cancer/testis-antigen genes by massively parallel signature sequencing

摘要: Massively parallel signature sequencing (MPSS) generates millions of short sequence tags corresponding to transcripts from a single RNA preparation. Most MPSS can be unambiguously assigned genes, thereby generating comprehensive expression profile the tissue origin. From comparison data 32 normal human tissues, we identified 1,056 genes that are predominantly expressed in testis. Further evaluation by using cancer cell lines and EST wide variety tumors 202 these as candidates for encoding cancer/testis (CT) antigens. Of tissues was assessed RT-PCR subset 166 intron-containing those with confirmed testis-predominant were further evaluated their 21 lines. Thus, 20 CT or CT-like identified, several exhibiting five more examined. One is member gene family designated CT45. The CT45 comprises six highly similar (>98% cDNA identity) clustered tandem within 125-kb region on Xq26.3. found frequently both lung specimens. analysis has resulted significant extension our knowledge antigens, leading discovery distinctive X-linked CT-antigen family.
