The purification of gametocytes of Plasmodium falciparum and P. yoelii nigeriensis by colloidal silica (Percoll) gradient centrifugation

摘要: Percoll gradient centrifugation has been applied to Plasmodium falciparam- and P. yoelii-infected blood allow rapid concentration of gametocytes. In the case cultured falciparum, stage IV V gametocytes were separated from immature II III forms concentrated initial gametocytaemias 1 3% an average greater than 80% a maximum 100% purity in fraction which collected at · 064/1 083 g/ml interface; rounded yoelli-infected mouse also this fraction. A much larger enrichment microgametocyte (average 160-fold) was recorded that 047/1 064 interface. The large increases gametocyte paralleled by similarly exflagellation activity, thus confirming viability both preparations.
