摘要: We report the cloning and sequence determination of mouse H19 gene. This gene is under genetic control two trans-acting loci in mouse, termed raf Rif. These determine adult basal inducible levels, respectively, mRNA, as well mRNA for alpha-fetoprotein. By elucidating structure we show that it unrelated to alpha-fetoprotein gene, therefore must have acquired its regulation by Rif independently. The also indicates has a very unusual structure. It composed five exons, 1307, 135, 119, 127 560 bp size, along with four small introns whose combined lengths are 270 bases. largest open reading frame sufficient encode protein approximately 14 kd, contained entirely within first large exon, 680 bases downstream cap site mRNA. Preceding translation initiation codon ATG codons, each which followed shortly thereafter terminator codons. rest encompasses all presumed be untranslated. That long 5' untranslated region may used regulate suggested from vitro studies. Experiments utilized tissue culture cell lines mesodermal lineage suggest activated early during muscle differentiation.
