Crosslinked poly(1-vinyl-2-pyrrolidinone) as a vitreous substitute

摘要: A hydrogel with a high water content was produced and tested as possible vitreous substitute. The polymer (poly[1-vinyl-2-pyrrolidinone]) (PVP) synthesized by free radical bulk polymerization of 1-vinyl-2-pyrrolidinone (VP) in the presence 0.25% divinyl glycol (DVG) crosslinking agent. fully hydrated polymer, containing about 98% water, clear, transparent, autoclavable, easily injected through small-gauge needle minimum fragmentation, without changes its optical properties. Dynamic mechanical analysis indicated covalently crosslinked elastic network both before after injection. resilience decreased being subjected to shear stress during injection process. cytotoxicity bioassay vitro, using cultured mouse (Balb/c-3T3) fibroblasts, showed cytostatic but not cytocidal effects. into cavity rabbits followed up 4 weeks. gel clinically well tolerated, however opacities body were observed following insertion gel. Histopathological examination revealed no adverse reactions retina, loose particles possibility biodegradation polymer. These results suggest potential use PVP hydrogels substitutes, provided that their is significant, matter should be further investigated. © 1996 John Wiley & Sons, Inc.
