The oncoprotein Bcl-3 directly transactivates through κB motifs via association with DNA-binding p50B homodimers

摘要: Abstract Bcl-3 is an IκB-related protein with ankyrin repeat motifs. Its gene located at a site of recurrent translocations in subset B cell chronic lymphocytic leukemias. associates tightly p50B (NFKB2, p52) homodimers cells, and together these proteins form ternary complex DNA κB sites. Such association functionally leads to novel potent transactivation through the motif: tethering via allows transactivate directly, while alone cannot. Transactivation mediated by requires two cooperating domains amino- carboxyterminal domain. localized nucleus, Bcl-3-p50B detected certain lymphoid cells. Our data reveal role for Bcl-3, distinct from that inhibitor IκB. The results have implications tumorigenesis.
