摘要: A simple and rapid isothermal absorptiometric assay for detection of viable microbes using the redox color indicator 2,6-dichlorophenolindophenol (DCPIP) was studied. The absorbance DCPIP decreased at 600 nm because a reaction occurring between surface membrane inversely proportional to cell density. found not only with bacteria, but also yeast mixture bacteria yeast. In this assay, influence light scattering absorption caused by microbial cells coexisting substances in sample excluded time difference method. required 10 min one incubation mixture, highly repeatable results from three consecutive measurements were obtained specific times 30 °C thermostable three-cuvette-stir system. Thus, density suspensions or growth medium successfully determined, practical lower limit food inspection 10⁴-10⁶ cfu/ml. Single-cell effects on reduction evaluated compared species. Consequently, is expected be useful tool measurement as preliminary Hazard Analysis Critical Control Point program.
