摘要: Tub4p is a novel tubulin found in Saccharomyces cerevisiae. It most resembles gamma-tubulin and, like it, localized to the yeast microtubule organizing centre, spindle pole body (SPB). In this paper we report identification of SPC98 as dosage-dependent suppressor conditional lethal tub4-1 allele. encodes an SPB component 98 kDa which identical previously described 90 protein. Strong overexpression toxic, causing cells arrest with large bud, defective structures, undivided nucleus and replicated DNA. The toxicity was relieved by co-overexpression TUB4. Further evidence for interaction between Spc98p came from synthetic spc98-1 alleles, suppression spc98-4 TUB4, binding two-hybrid system co-immunoprecipitation Spc98p. addition, Spc98-1p its system. We suggest model form complex involved organization SPB.
