摘要: We investigated the effects of treatment with tempol (an antioxidant) on vascular and metabolic dysfunction induced by a high-fat high-sucrose (HFHS) diet. Rats were randomized to receive an HFHS or chow diet without (1.5 mmol·(kg body mass)(-1)·day(-1)) for 4 weeks. Blood pressure, heart rate, blood flow measured in rats using intravascular catheters Doppler probes. Insulin sensitivity responses insulin assessed during euglycemic-hyperinsulinemic clamp. In-vitro studies performed evaluate reactivity endothelial inducible nitric oxide synthase (eNOS; iNOS) expression muscle tissues. Endothelin, nitrotyrosine, NAD(P)H oxidase expressions determined tissues, glucose transport activity transporter (GLUT4) examined muscles. Tempol was found prevent alterations sensitivity, activity, GLUT4 expression, reactivity, increases plasma insulin, rate noted untreated HFHS-fed rats. These associated increased levels eNOS but reductions endothelin, oxidase, iNOS expressions. Therefore, oxidative stress relatively short-term could contribute early development abnormalities
