The Development of an Efficient Multipurpose Bean Pod Mottle Virus Viral Vector Set for Foreign Gene Expression and RNA Silencing

摘要: Plant viral vectors are valuable tools for heterologous gene expression, and because of virus-induced silencing (VIGS), they also have important applications as reverse genetics function studies. Viral especially useful plants such soybean (Glycine max) that recalcitrant to transformation. Previously, two generations bean pod mottle virus (BPMV; genus Comovirus) been developed overexpressing genes in soybean. However, the design previous imposes constraints limit their utility. For example, VIGS target sequences must be expressed fusion proteins same reading frame polyprotein. This requirement limits open frames. Furthermore, expression multiple or simultaneous one another was not possible. To overcome these other issues, a new BPMV-based vector system facilitate variety studies well common (Phaseolus vulgaris). These designed foreign genes, insertion noncoding/antisense sequences, silencing. The green fluorescent protein phytoene desaturase shows marker gene-assisted is feasible. results demonstrate utility this BPMV set wide range bean, implications improvement plant virus-based systems.
