摘要: Under-agarose chemotaxis has been used previously to assess the ability of neutrophils respond gradients chemoattractant. We have adapted this assay chemotactic movement Dictyostelium amoebae in response folic acid. Troughs are instead wells increase area along which cells can be visualized and create a uniform front moving cells. Imaging transition zone where first encounter agarose, we find that move perpendicular gradient periodically manage squeeze under agarose up gradient. As exit troughs, their cross-sectional increases as become flattened. Three-dimensional reconstruction confocal optical sections through GFP-labeled demonstrates is due flattening Since locally deform deformed by it, concentration therefore its stiffness, should affect migrate. Consistent with hypothesis, 0.5% faster less flat than 2% agarose. Cells do not troughs 3% at all. Therefore, compare quantify different cell types or mutant lines restrictive environment.
