Evaluation of subchronic exposure to triclosan on hepatorenal and reproductive toxicities in prepubertal male rats.

摘要: Triclosan (TCS), a common antimicrobial ingredient, is present in many consumer products, including soaps, shampoos, and toothpaste. Owing to its widespread use, potential adverse effects on animals humans may arise from lifetime exposure, but data chronic prepubertal exposure of TCS are still lacking. The aim the study was investigate influence subchronic (0.25, 25, 250, or 750 mg/kg) target organ toxicity male rats. After daily administration rats by oral gavage for 60 d, significant reduction body weight relative weights liver, kidneys, testes, adrenal glands observed 750-mg/kg (high dose) group. Serum alanine aminotransferase aspartate activities as well levels blood urea nitrogen, creatinine were significantly increased at 750 mg/kg TCS. Further, (750 mg/kg) elevated protein expressions hepatic CYP2B1, RXR/PPAR, malondialdehyde. High-dose induced histological changes evidenced Bowman's space, occlusion tubular lumen, degeneration epithelial cells kidney. Tubular necrosis confirmed rise expression high mobility group box 1 renal protein. Daily sperm production diminished doses with marked inhibition androgen receptor expression. Our results indicated that excessively concentrations hepatorenal reproductive toxicities rats; however, biological relevance these findings questionable drug not encountered environment.
