Mass determination of the fatty acids released from tannin-stimulated rabbit alveolar macrophages

摘要: Previous studies with macrophages that had been prelabeled [14C]arachidonic acid (20∶4) have shown condensed tannin is a potent agonist for the release of arachidonic acid. However, it has not demonstrated percentage [14C]20∶4 accurately reflects metabolic activity endogenous 20∶4 pool. In order to measure mass relative total cellular pool, free fatty acids freshly isolated alveolar were derivatized fluorescent reagent, and then separated quantified by high-performance liquid chromatography. The amounts esterified measured gas chromatography methyl esters. Free levels compared those plus unesterified determine actual released each Tannin-stimulated reflected previously reported label but at slower rate much lower indicating incorporated into part more reactive specificity induced β-1,3-glucan, known release, was also examined. Both agonists promoted an increase in other acids. A comparison absolute increases indicated caused preferential 20∶4, whereas β-1,3-glucan evoked selective 16∶0.
