Conventional and Real-Time PCRs for Detection of Erwinia piriflorinigrans Allow Its Distinction from the Fire Blight Pathogen, Erwinia amylovora

摘要: Erwinia piriflorinigrans is a new pathogenic species of the bacterial genus that has been described recently in Spain. Accurate detection and identification E. are challenging because its symptoms on pear blossoms similar to those caused by amylovora, causal agent fire blight. Moreover, these two share phenotypic molecular characteristics. Two specific sensitive conventional real-time PCR protocols were developed identify detect differentiate it from amylovora other this genus. These based sequences plasmid pEPIR37, which present all strains analyzed. After stability was demonstrated, specificities confirmed amplification tested, whereas 304 closely related nonpathogenic microbiota trees not amplified. In sensitivity assays, 103 cells/ml extract detected spiked plant material or PCR, 102 DNA extracted PCR. The here succeeded detecting out 564 symptomatic asymptomatic naturally infected samples (flowers, cortex stem tissue, leaves, shoots, fruitlets), necrotic Pyracantha sp. blossoms, apple tissues with both piriflorinigrans. Therefore, tools can be used epidemiological studies will enhance our understanding life cycle different hosts interaction amylovora.
