摘要: Human antimicrobial RNases, which belong to the vertebrate RNase A superfamily and are secreted upon infection, display a wide spectrum of antipathogen activities. In this work, we examined antifungal activity eosinophil 3 skin-derived 7, two proteins expressed by innate cell types that directly involved in host defense against fungal infection. Candida albicans has been selected as suitable working model for testing activities toward eukaryotic pathogen. We explored distinct levels action both RNases on yeast combining viability membrane assays together with protein labeling confocal microscopy. Site-directed mutagenesis was applied ablate either active site or key anchoring region binding. This is first integrated study highlights RNases' dual mechanism action. Along an overall membrane-destabilization process, could internalize target cellular RNA. The data support contribution enzymatic proteins, can be envisaged templates development novel drugs. suggest human work multitasking provide line immune barrier.
