摘要: The cytokine-inducing activities of fungal polysaccharides were examined in human monocytes culture, with special reference to CD14 and Toll-like receptors (TLRs). Tumor necrosis factor alpha (TNF-alpha) production by was markedly induced a dose-dependent manner upon stimulation cell walls from Candida albicans mannan Saccharomyces cerevisiae C. albicans, although relatively high concentrations (10 100 microg/ml) stimulants required for activation as compared the lipopolysaccharide (LPS) (1 10 ng/ml). yeast form its wall fractions exhibited higher TNF-alpha than respective preparations hyphal form. Only slight S. glucan. triggered LPS purified mannans presence LPS-binding protein (LBP), these responses inhibited anti-CD14 anti-TLR4 antibodies, but not anti-TLR2 antibody. In contrast activity LPS, polymyxin B. These findings suggested that mannan-LBP complex is recognized on signaling through TLR4 leads proinflammatory cytokines similar LPS.
