摘要: Antiretroviral drugs are ineffective at treating viral infection in the brain because they cannot freely diffuse across blood-brain barrier (BBB). Therefore, HIV-1 replication persists central nervous system (CNS) and continues to augment neuropathogenesis process. Nanotechnology can play a pivotal role therapeutics as it increase drug solubility, enhance systemic bioavailability, same time offer multifunctionality. Moreover, following conjugation with transferrin (Tf), these drug-loaded nanoformulations permeate biological barriers such blood (BBB) via receptor mediated transport mechanism. In current study, we have stably incorporated antiviral drug, Saquinavir, within Tf-conjugated quantum rods (QRs), which novel nanoparticles unique optical properties. We evaluated transversing ability of QR-Tf-Saquinavir nanoformulation an vitro model BBB. addition, analyzed subsequent efficacy this targeted infected peripheral mononuclear cells (PBMCs), cultured on basolateral end BBB model. Our results show significant uptake by microvascular endothelial (BMVECs), constitute observed enhancement capability BBB, along marked decrease PBMCs. These observations indicate that deliver highlight potential treatment Neuro-AIDS other neurological disorders.
