摘要: The determination of the structural and functional contributions natural modified nucleosides to tRNA has been limited by lack an approach that can systematically incorporate units. We have produced a number oligonucleotide analogs, anticodon yeast tRNA(Phe) by, combining standard automated synthesis for major with specialty chemistries nucleosides. In this study, both naturally occurring unnatural nucleotides were placed in native contexts. Each was purified nucleoside composition determined validate chemistry. RNAs denatured analyzed determine van't Hoff thermodynamic parameters. Here, we report individual Cm, Gm, m1G, m5C, psi. addition m5m6U, m1psi, m3psi, introduced gain additional understanding physicochemical contribution psi m5C at atomic level. These oligonucleotides demonstrate modifications measurable loop global contributions.
