摘要: We have characterized a nuclease hypersensitive chromatin fraction from murine spermatozoa. Endogenous activity can be induced in mouse epididymal spermatozoa by appropriate stimuli and cause the localized degradation of chromosomal DNA. Based on these observations, we isolated regions released supernatant pelleted sperm cells, cloned Gel electrophoresis end-labelled DNA fragments showed typical nucleosomal distribution. Peripherally distributed nucleohistones were visualized immunofluorescence nuclei, histones identified western blot chromatin. Moreover, is enriched retroposon variety families. FISH analysis that nucleohistone co-localize are both peripherically nuclei In contrast, major satellite probe, used for control, co-localizes with highly condensed central region nuclei. The nuclear Ran RCC1 proteins also dorsal margin abundantly fraction. Together, results indicate fraction(s) features 'active' present spermatozoa, to cleavage, organized domains. These observations suggest domains identify genome which may functional during early embryogenesis.
