摘要: The protein products of the c-fos and c-jun proto-oncogenes (Fos Jun, respectively) form a heterodimeric complex that interacts with activator protein-1 (AP-1) binding site regulates gene transcription in response to extracellular stimuli. Protein dimerization is mediated primarily by coiled-coil-like structure termed leucine-zipper DNA occurs through regions each rich basic amino acids contact both strands AP-1 site. precise nature protein-DNA interaction unknown as studies concerned Fos Jun have relied on indirect methods investigate protein-protein-DNA interactions. Here we developed assay systems using fluorescence spectroscopy circular dichroism monitor directly. results indicate an altered conformation dimers increased alpha-helical content. These techniques may general application concerning transcriptional regulatory proteins specific target sequences.
