Rad51 suppresses gross chromosomal rearrangement at centromere in Schizosaccharomyces pombe

摘要: Centromere that plays a pivotal role in chromosome segregation is composed of repetitive elements many eukaryotes. Although chromosomal regions containing repeats are the hotspots rearrangements, little known about stability centromere repeats. Here, by using minichromosome has complete set sequences, we have developed fission yeast system to detect gross rearrangements (GCRs) occur spontaneously. Southern and comprehensive genome hybridization analyses rearranged chromosomes show two types GCRs: translocation between homologous formation isochromosomes which arm replaced copy other. Remarkably, all examined contain breakpoint repeats, showing produced rearrangement. Mutations Rad3 checkpoint kinase increase both GCRs. In contrast, deletion Rad51 recombinase preferentially elevates isochromosome formation. Chromatin immunoprecipitation analysis shows localizes at around S phase.
