摘要: Polyvalent bispecific antibodies were secreted by hybrid hybridoma cells when both parental clones expressed a naturally polymerizing immunoglobulin. Hybrid hybridomas made from IgA lambda 2 anti-trinitrophenyl (TNP) and kappa anti-phosphocholine (PC) polymeric that bound TNP PC. Some of the binding was dissociated PC under conditions mild reduction alkylation suggesting contained disulfide-linked monomers as well monomers. constructed anti-TNP IgM anti-ox erythrocyte bispecific, immunoglobulin mu-, alpha-, kappa-, 2-chains. The mu kappa-chains alpha- 2-chains alkylation, indicating had been incorporated into same to form heteropolymeric antibody molecule. Heterologous pairing alpha heavy chains in not detected. IgG3 or IgG1 parents co-secreted immunoglobulins, but these did heteropolymers exhibit heterologous chain pairing. These findings establish polyvalent, molecules can be produced express class only one parent does. produce immunoglobulins are sources high avidity may find number basic practical applications. provide useful tools for investigating situ determinants association regulation assembly secretion.
