Structural requirements for mutagenic activity of 2-acylaminofluorenes in the Salmonella test system

摘要: Abstract Twenty-nine 2-acylaminofluorenes were compared in the Salmonella typhymurium TA-1538 test system to determine effect of structural changes on mutagenic activity. All require S-9 liver fraction for Mutagenic activity was highest carbamate esters, ureas, α-haloacetylamides, and straight chain acylamides. Branching α-carbon, or addition unsaturation reduced Pretreatment C57BL 6 N mice with 3-methylcholanthrene increased (6–20 fold) all analogs fraction. No increase is observed from treated DBA 2 mice, indicating that correlates inducible aromatic hydrocarbon (benzo[a]pyrene) hydroxylase N-hydroxylase α-Naphthoflavone, an inhibitor N-hydroxylation 2-acetylaminofluorene vitro , inhibits these first step activation N-acylaminofluorenes into mutagens.
