摘要: A variety of therapeutic proteins have shown potential to treat central nervous system (CNS) disorders. Challenge deliver these protein molecules the brain is well known. Proteins administered through parenteral routes are often excluded from because their poor bioavailability and existence blood–brain barrier (BBB). Barriers also exist non-parenteral that bypass BBB. Several strategies promise in delivering brain. This review, first, describes physiology pathology BBB underscore rationale needs each strategy be applied. Second, major classes therapeutics along with some key factors affect delivery outcomes presented. Third, different administration (parenteral, intracerebroventricular intraparenchymal, intranasal intrathecal) discussed barriers CNS associated route. Finally, current involving chemical modification use particle-based carriers overviewed using examples literature our own work. Whereas most studies early stage, provide proof mechanism increased relevant models diseases, while few cases concept had been attained clinical studies. review will useful broad audience students, academicians industry professionals who consider critical issues aim developing studying effective systems for therapeutics.
