摘要: Hygromycin (hyg) at low doses (0.5–1.0 mg l−1) promoted somatic embryogenesis from apical sections of spinach lateral roots. The highest promoting effect on both the frequency regeneration and mean number embryos (SE) per explant was achieved 0.5 mg l−1 hyg. With increasing concentration hyg to 1 mg l−1, decreased, while SE remained significantly higher than in control (hyg-free medium). Complete inhibition started 7.5 mg l−1 Moreover, efficiently process secondary embryogenesis. Compared control, a 2.75-fold increase embryo (SSE) obtained hyg, increment still discernible 1.0 2.5 mg l−1 Both primary SSE explants became completely necrotic 12.5 mg l−1 Since attempts with direct selection 20 mg l−1 proved unsuccessful, results this study suggest that stepwise procedure is suitable, starting exploit transformed cells, then gradually for final selection. mortality above guarantees possibility escape during process. This will be useful efficiency transgenic plant following genetic transformation spinach.
