Glutamine protects against oxidative stress injury through inhibiting the activation of PI3K/Akt signaling pathway in parkinsonian cell model

摘要: Parkinson’s disease is a neurodegenerative disorder, and recent studies suggested that oxidative stress contributes to the degeneration of dopamine cell in disease. Glutamine also has positive role reducing damage. In this study, we hypothesized glutamine offers protection against injury 1-methyl-4-phenylpyridinium (MPP+)-induced model. MPP+ was used induce PD models PC12 cells classified into control, M0 (MPP+), G0 (glutamine), M0+G0 groups. CCK-8 AO/EB staining assays were examine proliferation apoptosis, respectively. Western blotting applied protein expression PI3K, P-Akt, Akt, P-mTOR, mTOR. We showed suppressed cytotoxicity induced by cells. decreased superoxide dismutase glutathione peroxidase activity increased malondialdehyde content, which restored glutamine. Moreover, mTOR, inhibited And antioxidant capacity on could be improved LY294002 IGF-1. These results suggest strengthens through inhibiting activation PI3K/Akt signaling pathway. The effects should investigated protective mechanism must explored future studies.
