摘要: How multiple chemoattractants cooperate in directing the migration of hematopoietic progenitor cells (HPC) for homing and peripheral blood mobilization has not yet been established. We report here behavior HPC under influence two different chemoattractants, stromal cell-derived factor (SDF)-1 steel (SLF), chemotactic nature bone marrow (BM) environment using a two-chamber vitro system. Various formulae were adopted to quantitate these effects. Based on quantitations, SDF-1 showed only activity, while SLF both chemokinetic activities factor-dependent MO7e cells. SLF, like SDF-1, attracted human from population CD34+ induced actin polymerization cooperated attracting cells, as well cord (CB) BM CD34+cells. A negative concentration gradient formed by presence upper chamber, potent inhibitory effects cell either lower synergistic mobilizing chamber this chemoattractant gradient. Plasma obtained aspirates, but CB or blood, strong an effect SDF-1–dependent migration. These experiments suggest that such with other unidentified may be involved cooperatively preventing spontaneous out BM.
