摘要: This paper is a review of strategies to introduce protein into the liquid medium Escherichia coli K-12 industrial production cells. The cell design are generally based on one two general mechanisms. first strategy involves two-stage translocation using active transporters in cytoplasmic membrane followed by passive transport through outer membrane. Passive achieved either external or internal destabilization E. structural components. latter can be transplantation destabilizing components (lysis proteins) that work permeabilization from interior cell, cells carrying mutations also chemical, mechanical, enzymatic directed outside cell. second realized proteins capable over both membranes. secretion mechanisms pathogenic as well other species. process dependent environmental conditions and must take account changes physical parameters, design, influx limiting carbon source fed-batch cultivation.
