摘要: The function of the Src-family kinases (SFKs) Lck and Fyn in T cells has been intensively studied over past 15 years. Animal models cell line studies both indicate a critical role for proximal T-cell antigen receptor (TCR) signal transduction. Recruited SFKs phosphorylate TCR ITAMs (immunoreceptor tyrosine-based activation motifs) CD3 ζ chains, which then serve as docking sites Syk-family kinases. activate recruited kinase. are spatially segregated membranes due to differential lipid raft localization, may undergo sequential activation. In addition CD4 CD8 coreceptors, recently described adaptor, Unc119, link TCR. TCR-based signaling required at several stages development, including least pre-TCR signaling, selection, peripheral maintenance naive cells, lymphopenia-induced proliferation. each these signals, seems be major contributor.
