摘要: A quantitative assay for infective variola virus particles was developed which is based on the enumeration of cells containing fluorescent viral antigen after infection McCoy cell monolayers. The direct fluorescent-antibody technique employed to stain cells. efficiency adsorption markedly enhanced by centrifugation inoculum onto monolayers at 500 x g 15 min. By this procedure, a proportionality obtained between number and volume inoculum. Observations sequential development within counts showed that optimal time enumerating an incubation period 16 20 hr. linear function existed concentration cell-infecting units. Fluorescent were distributed randomly in infected cover slip demonstrated be highly sensitive, precise, reproducible.
