Isolation and Identification of Rhizospheric Bacteria in Sugarcane (Saccharum spp. L.) Cultivated on Acrisols and Ferrasols of Dong Nai Province, the Southeast of Vietnam

摘要: Nitrogen-fixing and phosphate-solubilizing bacterial diversity population dynamics in the Acrisol Ferrasol rhizosphere of sugarcane grown Dong Nai province, eastern South Vietnam was studied. Soil samples were taken three districts (sites) this region. Physical chemical characteristics soil total nitrogen-fixing bacteria counts determined by drop plate count method together with 16S rRNA gene fragments amplified from DNA using eubacterial universal primers (27F 1492R). A 31 isolates isolated on two media (Burk’s N-free NBRIP) all them have ability nitrogen fixation phosphate solubilization IAA biosynthesis. Population correlated N concentration organic matter content closely (P<0.05). The sequences selected (12 isolates) showed high degrees similarity to those GenBank references strains (between 98% 99%). From 12 isolates, 7 belonged Bacilli, while 2 Beta-Proteobacteria, 1 Acidobacteria Bacteroides. Based Pi value (nucleotide diversity), Bacilli group had highest theta Theta values (persequence) S SNP for polymorphism calculated each comparison groups. these results that (Bacillus megaterium B6 (Ferrasols) Sphingomonas sp. P14 (Acrisols)) revealed promising candidates multiple beneficial they potential application as inoculants adapted poor soils tree because are not only famous but also safety agricultural sustainable.
