The ubiquitin-specific protease USP8 is critical for the development and homeostasis of T cells

摘要: The modification of proteins by ubiquitin has a major role in cells the immune system and is counteracted various deubiquitinating enzymes (DUBs) with poorly defined functions. Here we identified ubiquitin-specific protease USP8 as regulatory component T cell antigen receptor (TCR) signalosome that interacted adaptor Gads molecule 14-3-3β. Caspase-dependent processing occurred after stimulation TCR. cell-specific deletion mice revealed was essential for thymocyte maturation upregulation gene encoding cytokine IL-7Rα mediated transcription factor Foxo1. Mice deficiency developed colitis promoted disturbed homeostasis, predominance CD8(+) γδ intestine impaired function. Collectively, our data reveal an unexpected immunomodulatory DUB cells.
