摘要: Up to 1% of the population have mild bleeding disorders, but these remain poorly characterized, particularly with regard roles platelets. We compared usefulness Optimul, a 96-well plate-based assay 7 distinct pathways platelet activation characterize inherited defects in comparison light transmission aggregometry (LTA). Using Optimul and LTA, concentration-response curves were generated for arachidonic acid, ADP, collagen, epinephrine, Thrombin receptor activating-peptide, U46619, ristocetin samples from (1) healthy volunteers (n = 50), (2) treated antiplatelet agents vitro 10), (3) patients unknown origin 65). The assays gave concordant results 82% cases (κ 0.62, P < .0001). Normal function predictive (sensitivity, 94%; negative value, 91%), whereas positive result was not always substantiated by LTA (specificity, 67%; 77%). significantly more sensitive at characterizing thromboxane pathway, which presented normal responses LTA. is defects, could be used as rapid screening presenting symptoms, detects changes readily than This trial registered www.isrctn.org #ISRCTN 77951167.
