Molecular cloning, immunohistochemical localization, characterization and expression analysis of caspase-9 from the purse red common carp (Cyprinus carpio) exposed to cadmium

摘要: Caspase-9, the essential initiator caspase is believed to play a central role in mitochondria-mediated apoptosis signaling. In this study, we isolated caspase-9 gene from common carp, one of most important industrial aquatic animals China using rapid amplification cDNA ends (RACE). The deduced amino acid sequence caspase-9, composed 436 acids, showed approximately 47.6% identity and 64.7% similarity human caspase-9. It also possessed conserved caspase-associated recruitment domain (CARD), large subunit small subunit. Phylogenetic analysis clearly demonstrated that formed clade with cyprinid fish Real-time quantitative PCR revealed transcripts were not significantly increased kidney after exposure cadmium (Cd). Whereas cleaved fragments detected Western blot same Cd treatment condition. Furthermore, result immunohistochemical detection immunoreactivities predominantly limited cytoplasm renal tubular epithelial cells no remarkable changes immunopositive staining observed treatment. Accordingly, results signify may an induced apoptosis.
