摘要: Abstract Glycan structure determination serves as an essential step for the thorough investigation of and function protein. Currently, appropriate sample preparation followed by tandem mass spectrometry has emerged dominant technique characterization glycans glycopeptides. Although extensive efforts have been made to development computational approaches automated interpretation glycopeptide spectra, previously appeared methods lack a reasonable quality control strategy statistical validation reported results. In this manuscript, we introduced novel method that constructed decoy glycan database based on structures in target database, searched experimental spectra against both databases find best matched glycans. Specifically, two-layer scoring scheme calculating normalized matching score is applied search procedure which enables unbiased ranking Experimental analysis showed our proposed can report more with high confidence compared previous approaches.
