摘要: A 16.5-kb BamHI fragment of the Cellvibrio mixtus chromosome was found to direct carboxymethylcellulase, xylanase, and avicel hydrolysis. Two closely linked genes were subcloned from this insert. The gene, cmcI, cloned as a 2.7-kb expressed in Escherichia coli. It encoded an enzyme approximately 74 kDa which degraded carboxymethylcellulose xylan but did not attack microcrystalline cellulose substrate avicel. second cellulase capable degrading avicel, by exoI, 5.5 kb downstream cmcI. translation products 53.7 51.5 produced E. coli strains expressing exoI. Northern analysis total mRNA C. grown on with probe generated showed that cmcI exoI cotranscribed operon.
