摘要: Pluripotency and self-renewing ability of embryonic stem (ES) cells are regulated by several transcription factors, including Oct3/4, Sox2, Kruppel-like factor 4 (Klf4), c-Myc. These factors reprogram somatic into induced pluripotent (iPS) cells. Zinc finger protein (Zfp) 296 has been reported to enhance iPS cell formation. Here we found that Zfp296 interacts with Klf4. A maltose-binding pull-down assay demonstrated Klf4 binds the 158-483 amino acid region, DNA-binding domain (DBD). quantitative reverse transcription-polymerase chain reaction analysis revealed expression decreased during differentiation E14 ZHBTc4 ES We also green fluorescent protein-labeled were localized nucleus. Because bound DBD, next examined influence on activity. biotin DNA showed Lefty1 promoter binding activity was sustained even in presence Zfp296. In contrast, a reporter activated Klf4, enhanced repressed findings suggest is functional regulator
