Concentration x time methotrexate via a subcutaneous reservoir: a less toxic regimen for intraventricular chemotherapy of central nervous system neoplasms

摘要: Neurotoxicity associated with intrathecal methotrexate therapy has been shown to correlate elevated concentrations of the drug in cerebrospinal fluid as well total cumulative dosage. In our study 19 patients meningeal leukemia were randomized receive courses intraventricular via an Ommaya reservoir consisting either single injections 12 mg/sq m/dose or a low-dose “concentration x time” (C T) schedule 1 mg every hr for 3 days. There no significant differences between two treatment groups rate remission induction, number relapses, durations remission. The mean (+/- SD) dose was 66 +/- 41 m C T group and 173 64 (p less than 0.005). Neurologic toxicity occurred one eight seven ten 0.05). These observations suggest that dosage is neurotoxic equally effective central nervous system leukemia.
