Electrokinetic concentration on a microfluidic chip using polyelectrolytic gel plugs for small molecule immunoassay

摘要: Abstract We have devised a new method to enhance the sensitivity of competitive immunoassay using electrokinetic concentration near pair highly charge-selective polymer [poly-2-acrylamido-2-methyl-1-propanesulfonic acid (pAMPSA)] plugs on microfluidic chip. The polyelectrolytic gel, which was photopolymerized in channel network, served as effective extractor sophisticatedly control ion distribution. In this system, fluorescent indicators magnetic microbeads dispersed sample were spontaneously displaced by unlabeled target molecules and then electrokinetically preconcentrated single spot locally detected laser-induced fluorescence. As proof-of-concept, displacement assay 1 nM biotin conducted observe ca. 2000-fold enrichment within 3 min. addition sensitive assessment small molecules, proposed system also showed good selectivity for analogs such biocytin, 2-iminobiotin, desthiobiotin.
