Verteporfin-Loaded Lipid Nanoparticles Improve Ovarian Cancer Photodynamic Therapy In Vitro and In Vivo.

摘要: Advanced ovarian cancer is the most lethal gynecological cancer, with a high rate of chemoresistance and relapse. Photodynamic therapy offers new prospects for treatment, but current photosensitizers lack tumor specificity, resulting in low efficacy significant side-effects. In present work, clinically approved photosensitizer verteporfin was encapsulated within nanostructured lipid carriers (NLC) targeted photodynamic cancer. Cellular uptake phototoxicity free NLC-verteporfin were studied vitro human cell lines cultured 2D 3D-spheroids, biodistribution evaluated vivo mice. Both molecules internalized cells strongly inhibited viability when exposed to laser light only. pharmacokinetic studies evidenced long circulation time NLC associated efficient uptake. Administration 2 mg.kg-1 induced severe phototoxic adverse effects leading death 5 out 8 contrast, exposure tumors after intravenous administration (8 mg.kg-1) significantly growth without visible toxicity. thus led vectorization site protection from side-effects, providing promising therapeutic
