摘要: Human pluripotent stem cells (PSCs) represent an opportunity to study human development in vitro, model diseases a dish, screen drugs as well provide unlimited and ethically unimpeded source of therapeutic cells. Cortical GABAergic interneurons, which are generated from Medial Ganglionic Eminence (MGE) Caudal (CGE) during embryonic development, regulate cortical neural networks by providing inhibitory inputs. Their malfunction, resulting failure intricately circuit balance, has been implicated brain diseases, such schizophrenia, autism epilepsy. In this study, using combinatorial temporal modulation developmentally relevant dorsoventral rostrocaudal signaling pathways, we efficiently MGE vs. CGE PSCs, predominantly generate Parvalbumin-expressing or Somatostatin-expressing interneurons Calretinin-expressing respectively. Efficient generation specific differentiated progenies hPSCs shown will be pivotal step realize the full potential for regenerative medicine, developmental studies, disease modeling, bioassay, drug screening.
