摘要: Somatic Brassica napus (+) Arabidopsis thaliana hybrids with a cytoplasmic male sterility (CMS)-inducing cytoplasm were screened for fertility-restored plants. One line was selected and recurrently backcrossed the maintainer line, B. napus, resulting in fertile/sterile segregating populations. Restriction fragment length polymorphism mapping showed co-segregation of A. chromosome (chr) III markers fertility trait. As it not possible to stabilise trait via selfings, dihaploidisation strategy assessed. Ninety haploid plants regenerated analysed numerous simple sequence (SSLP) markers. Markers covering both arms chr present two plants, whereas no DNA could be detected other Following colchicine-induced doubling only these produced fertile offspring. In one lines, however, thaliana-specific lost subsequent generations. The remained after repeated selfings. Using genomic situ hybridisation (GISH) we able demonstrate that this latter possessed disomic addition chromosome. restored comparable respect flower morphology, but petals stamens slightly reduced size. homeotic conversion pistil-like structures, which is typical CMS reversed, normal appearance viable pollen appeared. Flowering time as line—in lines delayed comparison line. introgressed also contributes several pleiotropic effects, such leaf crinkling shorter stems. ability restore through introgression nuclear genes from main donor species indicates system mainly due napus/A. alloplasmic incompatibility mitochondrial rearrangements. Further exploitation material discussed.
