An essential role for the Cdc6 protein in forming the pre-replicative complexes of budding yeast

摘要: ORIGINS of DNA replication in Saccharomyces cerevisiae are bound by two protein complexes during the cell cycle1,2. Post-replicative closely resemble those generated vitro purified origin recognition complex (ORC)1,3–5, which is essential for vivo6–11. Pre-replicative (pre-RCs) characterized an extended region nuelease protection overlapping ORC footprint1. We show here that Cdc6 (Cdc6p), necessary firing vivo12–16, establishment and maintenance pre-RCs, suggesting it a component these complexes. Without Cdc6p, G1 origins post-replicative origins, providing evidence also pre-RCs. These results suggest pre-RCs play role initiating support two-step mechanism assembly functional initiation
