摘要: Chemical castration involves administration of drugs to prevent pathological sexual behavior, reduce abnormal drive and treat hormone-dependent cancers. Various have been used for chemical castration; however, substantial interindividual variability side effects are often observed. In this study, we proposed a useful monitoring method the application agents using ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS-MS). Testosterone, cyproterone acetate, medroxyprogesterone, goserelin leuprolide acetate triptorelin were analyzed by UPLC-MS-MS. The target extracted from serum samples double protein precipitation methanol. Testosterone-1,2-d2 buserelin as internal standards. Parameters analytical performance evaluated, including imprecision, linearity, ion suppression detection capabilities. Testosterone measurements compared with results immunoassays. Serum specimens 51 subjects who underwent analyzed. All testosterone well separated our method. was essentially free potential interferences suppression. Within-run between-run imprecision values <15%. lower limits quantification 0.125 0.5-1.0 ng/mL other drugs, respectively. Good correlations pre-existing immunoassays measurement Sera androgen deprivation therapy showed variable levels drugs. We successfully developed UPLC-MS-MS-based castration. generally acceptable. This may provide novel strategy enhance expected while reducing unwanted effects.
