摘要: Discrimination of Alu insertions is a useful tool for geographic ancestry analysis, and usually performed by element amplification agarose gel electrophoresis. Here, we have developed new fluorescence-based method multiple genotyping in forensic identification. Allele frequencies were determined 70 Japanese individuals, selected 30 polymorphic insertions. Three primers designed each locus to discriminate alleles using the 3-6 bp differences amplicon sizes. Furthermore, classified loci into three different sets, PCR set provided 10 fragments ranging from 50 137 bp. Based on population data, probability incorrectly assigning match was 3.7×10(-13). independent amplifications subsequent capillary electrophoresis enabled sensitive small amounts DNA, indicating that this suitable identifying individuals ethnicity.
